Humanin and longevity signaling: where is the human data?

Humanin keeps showing up in longevity discussions for its cytoprotective and neuroprotective signaling. From what I can find, though, the evidence is entirely preclinical and mechanistic. Is there any human efficacy data I am missing?

If not, this seems like a clear Tier 4 situation: a compelling biological story with essentially no controlled human results behind the aging claims.

You are not missing anything. There is no human efficacy data for humanin in aging endpoints; the evidence is cell and animal models plus mechanistic work. For the longevity claims specifically, Tier 4 is the right label. It is a compelling biological story, but a story is not a controlled human result.
